Job Description Summary:

The Associate Scientist I, Multiomics Analysis (Imaging) will use cutting edge analytical techniques to determine the localization of biomarkers in situ as well as their abundance in tissues and biofluids. The candidate will join a diverse team responsible for generating data to help elucidate our drug's mechanism of action and the underlying disease pathology in both genetic and pharmacologically induced pre-clinical models of human disease. This position will aid in the group's efforts to provide evidence of target engagement across the PTC Therapeutics pipeline with a particular focus on neurodegenerative programs, ferroptosis, mitochondrial dysfunction, and metabolic disorders. The incumbent will be responsible for communicating experimental results to his/her supervisor and working cross-functionally across multiple departments.
This position will adhere to relevant regulatory requirements and company Standard Operating Procedures (SOPs) as appropriate.Job Description:

ESSENTIAL FUNCTIONS

Primary duties/responsibilities:

  • Execute sample preparation procedures for cells, biofluids, and tissues to conduct multiomics analyses from a single specimen.
  • Run quantitative immunoassays for both human and rodent biomarkers using MSD, Quanterix, and Ella platforms.
  • Perform LC-MS/MS to quantitate endogenous biomarkers using MRM analyses.
  • Assist with multi-modal imaging experiments (i.e. imaging mass spectrometry, IF, IHC) to visualize biomarker distribution in tissue sections.
  • Prepare/maintain reagents and stock solutions for biomarker quantitation and imaging experiments.
  • Abides by all safety and regulatory guidelines. Maintains a detailed laboratory notebook.
  • Communicates effectively with peers and presents data at weekly group meetings.
  • Demonstrates compliance with corporate, department, and job-specific requirements.
  • Performs other tasks and assignments as needed and specified by management.

KNOWLEDGE/SKILLS/ABILITIES REQUIRED

  • Experience quantitating protein biomarkers using automated "capture/reporter" instruments such as: MSD, Quanterix, Ella, or Nanostring.
  • Experience with brightfield and fluorescence microscopes (preferrable from Zeiss).
  • Extensive hands-on experience using mass spectrometers (e.g. Sciex, Bruker, etc.) with various mass analyzers (i.e. QqQ, Orbitrap, Q-ToF, etc.) for targeted and global biomarker analysis.
  • Prior use of UHPLC systems for chromatographic separation of metabolites, lipids, or proteins.
  • Wet-lab experience in sample processing techniques enabling the isolation, enrichment, and purification of multiple analytes in biological matrices.

* Minimum level of education and years of relevant work experience.

  • Bachelor's degree in Analytical Chemistry or Biochemistry with 2+ years of relevant experience or Master's degree in Analytical Chemistry or Biochemistry with 0-1 year of relevant experience

* Special knowledge or skills needed and/or licenses or certificates required.

  • Detailed knowledge of tissue staining protocols used for IHC and IF.
  • Familiarity with cryo-sectioning.
  • Knowledgeable of ELISA based protein quantitation techniques.
  • Prior use of statistical software to quantitate and analyze biomarker datasets.
  • Excellent verbal and written communication.
  • Detail-oriented worker with superb organizational and time management skills.
  • Analytical thinker with excellent problem-solving skills.

Pay Range: $75,000 - $105,000

The base salary offered will be contingent on assessment of candidate education level, background, and experience relative to the requirements of the position they are being considered for, as well as review of internal equity.

In addition to base salary, PTC employees are also eligible for short- and long-term incentives.All eligible employees may also enroll in PTC's medical, dental, vision, and retirement savings plans.
